Depends on the context of the site that is selling the t-shirt. It could be considered a satirical joke, but it would be a very bad one.

It's a joke if people find it funny. Whether or not it's intended that way. I for one find the concept of a 'Proud Republican' to be hilarious, so I guess that makes the answer to your question a Yes.

No, the Elephant has long been the symbol of the Republican Party. The Donkey is the same thing, for the Democratic Party. I really don't think it's a joke, but in a way that makes it funnier.

I would say it depends on the viewer.

Myself, if I saw anyone wearing either shirt, I would find it humorous. Showcasing your pride in a political agenda via a t-shirt is rather funny to me.

PETA would be even better...
