That sounds like The Forgotten Capital with being inaccessible because of the Sleeping Forest which only awakes with the Moon Harp and Kujata as Elemental Lord.
It could also be the Round Island Cave, but for the Forgotten Capital the text is more fitting. Except you don't mean real trees, hm ...

I'm gonna take another guess but we might need a clue soon. When I say "we" obviously I mean "I" at the moment!
Is it the Temple of the Ancients? Again, I don't think it really fits but it's inaccessible without the Keystone, it has more than one 'guardian' and it's surrounded by trees.
Man, that was a pretty lame guess actually... awaiting the inevitable rejection! Hopefully someone will come along soon who is better at this than me.

Hint: The Forgotten Capital is the closest guess so far, in terms of storyline progression.
The closest? You mean the Cave which cannot be entered where a way leads down to a vine which cannot be climbed up? Or short, the thing that is called the "Cave of the Ancients"? That's an obvious one as well then. I didn't take that inaccessible and all that literally.
