Inuyasha and Kagome.
You can't argue with the music.
At the time I first watched it, Inuyasha was probably the most "romance oriented" thing I had ever encountered. There are undoubtedly much better love stories out there as even I acknowledge Ms. Takahashi was a bit repetitive. Also I can see why some people hate the "obvious love story" story. And yet I still find their relationship extremely touching and beautiful. Nostalgia? Yes, that is most assuredly a factor. And yet I feel they had several very powerful moments throughout the series.
Nephrite & Molly/Naru - Sailor Moon
Train & Saya - Black Cat
Junta, Karin & Ami - DNAČ
Itroutaka & Iori - I''s
Keitaro & Naru - Love Hina
And we all know we love it: Titanic.
I love the story of Sarah and Chuck in Chuck. Just... amazing!
David and Elise from The Adjustment Bureau. Matt Damon and Emily Blunt's chemistry was so goddamn good in that movie.
1Q84. Such a great book. I'm normally not into sappy love story crap but Murakami is a smurfing amazing writer. He wrote a love story in a way that's not retarded but actually interesting.
Cowboy Bebop - Spike and Juliet. Such a tragic tale, and one of the greatest endings of all time.
Trigun - Wolfwood and Millie. Also ends in sadness.
Rocky Horror Picture Show - Frank and everyone. Also kind of a sad ending.
Teen Titans - Robin and Starfire, as well as Beast Boy and Terra, the later of which was another sad ending to it.
...Yeah, I have a thing for love stories that end saddly. :p
Edit: Add in Seras and Pip from Hellsing Ultimate.
i love the story of van helsing...
Julia! But yes, I looove them.
Ben Wyatt and Leslie Knope are at the top of my list right now, as you can see all over the face of babe's profile (Parks and Rec). Also Andy and April!
Jim Halpert and Pam Beesley, though I like them better pre-marriage (The Office).
I'm trying to think of chick flicks but I'm coming up short, same with romantic books. As much as romantic tension between characters really makes me squee, I think that I kindof prefer most of my stories to be without being centered around it.
Why the hell did I write Juliet? XD
Cause you were thinking of Romeo and Juliet and wanted to name them, too
I didn't like Romeo and Juliet. XD
I'll chalk it up to it was after my vein collapsed and smurfed with the blood flow to my brain. ;P
MARS is still my favorite manga.

Also Pride and Prejudice. Especially the movie.

Here are some examples which crossed my mind:
I enjoyed the love story of Eureka and Renton (spoilers in the video) in Eureka Seven. Even if it dragged out a bit, I was impressed by how well they developed the romance over the episodes and how ultimately had such a satisfying ending despite all the hardships they had to go through to find the road to acceptance. :)
Then there's the destructive romance of Johnny and Gilda from Gilda with this memorable exchange which the whole movie has been setting up. Great stuff from Glenn Ford and Rita Hayworth, and the film is definitely worth checking out for any fan of romance or film noir.
Vanyel and Tylendel from Mercedes Lackey's The Last Herald-Mage series. smurfing makes me depressed and feel like I will never find love.
Miaka and Tahome from Fushigi Yuugi. I was yelling at the TV towards the end and threatening it if they did not end up together.
Merlin and Arthur from Merlin fanfiction. Gets me in the feels every time.
Christian and Satine, Aragorn and Eowyn, Shrek and Fiona, Giselle and Robert, Peter Parker and Gwen Stacy, Anna and Declan, Harry and Sally, Lloyd and Diane, Hermia and Lysander, Nick and Darcy (particularly at the end when he claims he came clean to Darcy as an act of heroism but realizes he is actually in need of saving)
Norwegian Wood
Jude the Obscure
Lost in Translation (this counts to me)
and for something less depressing
trout, can't think of anything lol
Anyone who says Romeo and Juliet is missing the point. :p
Anyway, Silver Linings Playbook had an excellent romance to it. Right now, that's one of my favorites.
Frodo and Sam
Sir Gawain and Lady Ragnelle.
My favorite story of King Arthur's knights as well.
