You can go back. The map resets everytime you enter a new game and the leg will be there again and again.

If you don't have a Leg in your inventory, a new one will drop from Wirt's body in Single-Player. I'm pretty sure that even if you do have one, a new one will drop anyways; you just won't be able to pick it up. As Cim said, though Tristram's map doesn't vary, you can still revisit it; for example, Griswold will try to kill you every single time.
And before mentioning only being able to do the Cow level once, this is also untrue. As long as you have killed Baal and have failed to kill the Cow King, you can create a Portal. In multiplayer, as long as you are unpartied and not in the Cow level at the time of the Cow King's death, you will still have the ability to create the Cow level. The truthful conditions for extermination are actually something in between, but as far as I'm concerned, BOTH conditions must be true to preserve your order of steak, five seconds rare. Alternatively, you can make someone else do it and kill the King as many times as you want. You just won't be able to make it yourself.
Also, Hostilities will prevent you from taking the Portal, unless you are already in the Cow level, and have spare TP scrolls.
