You've got the first one right. The second one could arguably be right, but there are two more important meanings to the line.
Correct! Reasoning as follows:
I am endless toil - He works constantly, hammers away on his sign endlessly until...
Interrupted - down and up - Vivi trips, making him miss. He climbs down the ladder, then back up.
No comedian I - This is a reference to the signmaker's name being Dante, like the author of Divine Comedy. But sign maker Dante is no comedian - instead he gets angry at Vivi's little comical tripping.
Yet it somehow helps - But it seems that it helped, because though he worked eternally before...
Thrice more I strike, then punch out - When he returns, he only strikes the sign three more times. Then he "punches out", ie. leaves work.
Do I smell a rat? - A reference to Puck who arrives on the scene, who apart from being a rat is also up to no good, which the phrase suggests.
