I would like a real for serious Deadpool movie. Not that trout they tried to pull in XMEN though.

An actual adaption of Rogue as depicted in the comics would be a start.
Wonder Woman, but it will never happen because the WB are being stupid about it.
Edit: haha, I for some reason thought this was just about the women. I second the Deadpool movie. The game was awesome, do something like that.
Azrael. What they should have done for the third Nolan Batman movie was have Jean-Paul Valley step in as Azrael Batman because it really hammers home the steely, unbending will of Batman when you see him contrasted against another person who tries to wear the mantle.
Wonder Woman!
They're including her in the new Batman and Superman movie, and I think it's to test the waters and get a reaction for her onscreen character. I think we'll see her get her own film by 2016, though I am not pleased with the actress they chose for the role.
This is the best fan trailer I've ever seen in my life, and I wish they would use this lady for it if they chose to make a movie of WW. Watching this gives me chills and I think the potential for this trailer to be elaborated by a team of seasoned pros behind the works would make for an amazing film.
Last I heard she hasn't gotten her own movie, despite people saying they'd be interested in one, is because WB claims people don't want to see a Wonder Woman movie and gives us repeated Batman and Superman films instead. First I heard of her being included in the Batman/Superman movie.

Whether or not WB are being buttheads about it now, I think we'll see a film of her own. She's a top female superhero, if not the top female superhero, the likes of which not many movies are made about. The only one I can think for the top of my head really is Elektra, although I'm sure there are more. edit: oh and the travesty of catwoman and halle barry. They'll make one of Wonder Woman eventually.
Black Widow is too much of a side character to warrant her own film, I think, although the folks behind The Avengers movies might want to capitalize on that entire team and their own films.
I can't think of too many other prominent female superheroes who stand in the spotlight of their own achievements (because I am not overly familiar with the world of superheroes), but I would love a Zatanna Zatara film
I dislike how ridiculously sexualized her appearance generally is, though. This is like the one photo I found where her breasts weren't coming out of her vest and her suit wasn't essentially turned into a thong over fishnets.
Really, if I was for some reason in charge of making a DC Cinema Universe ideally I would want to start off with a Superman, Batman and Wonder Woman movie since they're the trinity of DC comics. I'd want to do indiviual films of Aqua Man, Green Lantern and Flash too, though Flash could be tricky. Only one I wouldn't give his own film would be Martian Manhunter since he's more of an observer type character. He'd be better off used to tie all the movies together leading to the Justice League payoff.
I feel like I'm forgetting someone.
The first DC film starring a female superhero should obviously star Power Girl.
But who could "fill" that role?
Wonder Woman and She-Hulk. I wouldn't mind seeing a Black Widow film either although I know people have had wildly varying reactions to her character.
Among male superheroes, I agree Deadpool would be a pretty good choice.
You'll be disappointed then. Word has it that Wonder Woman and some others are getting cameos in Superman/Batman because they're planning a Justice League movie next. Couldn't be more annoyed by that though. Not that I'm not curious about a Justice League movie, I just don't like the fact that WB apparently has no faith in any hero being able to carry a movie aside from Batman and Superman. There are so many interesting and deserving heroes in the DC stable but they're afraid to use them.
I don't think I'd have any interest in a Just League movie. I only barely had interest in the Avengers because a) the reviews were outstanding and b) Joss Whedon (and c) Chris Evans).
This is why I like the people behind the Marvel movies, they were willing to take a chance on Iron Man, Hulk after a failed first time, Thor and Captain America instead of just being like, "Oh, well, the only movies that will sell are Spider-Man and X-Men, lol!"
