O.K my question is a two-parter-first and this might sound strange, but in the cloister of trials thing is there any reason why I shouldn't go to the first level before I reach the 20 level increments, and is there a strategy to fighting the Mega-Tonberries?

not too sure about your first question, but i guess if you return to the first level (cloister 0) before reaching the great cloisters, then you have to go through all the levels again.
as for your second question, the best strategy against mega tonberries is to avoid them. unless you're trying to fight them on purpose, then trigger happy + catnip works for me. firststrike comes in handy so it's better if you have sprint shoes equipped or use the highroad winds gg (they say mounted assault gg also has firststrike but so far i haven't gotten this gg yet)
No, if you just step on the pad on the surface it'll let you go back to the last cloister reached. I just wanted to know if it counted for 100% completion if I did that.

Yeah, if you complete Via Infinato it adds to your completed score.
