It does seem to help imo. There are some solid numbers out there somewhere. A program was written to input your synth results and several variables. You could do this over a long period of time and compile what variables seemed to have what % effect on your results. People who have kept track have results that usually line up with synthesis theory.
My wife is a high level Woodworker and she definitely takes heed to direction, day, and moon phases. There is a very noticeable affect on HQ results as a result of these things. Moon phases seems to play the biggest part.
Having taken 4 crafts to 60 myself, I use diretional crafting to speed up my skill ups. When I far below the cap I will face the successful NQ direction and as I get closer I will either face a neutral direction or the HQ direction (which fails more often, especially under the cap).
Most directional crafting theories are based on a combination of strength/weakness of elements to one another (absolute fact) and the constellation chart you find by speaking to an NPC in (I think) Windurst Waters.
A 100+3 Bonecrafter friend of mine on Fenrir is easily one of the most successful bonecrafters. He believes absolutely in directional crafting and pays very close attention to all variables involved. He's made several Dragon Harness +1s among a cadre of other incredible HQ.
