Actually, mixing up "your" with "you're" IS pretty serious.

It's another case if you go general like "it ain't no good" or "this don't appeal to me" and the like, but "your" and "you're" are two COMPLETELY different types of words!

"Give me the pens over there, please."
"Give me the penis over there, please."
